Introduction：

产品介绍：

SC Broth是由酵母氮源（YNB）与缺陷型氨基酸混合物（DO supplement）混合而成，补加葡萄糖后即为SD Broth。培养基中的葡萄糖在高温灭菌过程中会发生美拉德反应出现碳化情况，培养基变黄棕色，严重甚至变成棕黑色。对于碳源碳化敏感的酵母菌种，碳源需要单独过滤除菌再加入灭菌后的SC Broth培养基中。葡萄糖的微量碳化对经常应用于酵母杂交实验中的酿酒酵母菌种影响并不大，没有特别要求一般建议直接选用SD Broth。该产品如需制作平板，按20 g/L添加琼脂，高压灭菌后冷却至60℃左右加入预热的碳源（过滤除菌），混匀倒板即可。

 

配制方法：

1. 在900 mL去离子水中加入8 g的SC Broth，搅拌溶解。

2. 无需调整pH值。121℃高压灭菌15分钟。

3. 4℃冰箱避光储存已经灭菌的液体培养基。

4. 使用前加入100 mL已过滤除菌的20%葡萄糖溶液，或者其它碳源。
